The processed swab sample is applied to the strip's sample pad.

HRV antigen (if present) binds to gold-labeled mouse anti-HRV antibodies in the conjugate pad, forming an antigen-antibody-gold complex.

The complex migrates along the nitrocellulose membrane via capillary action.

 A second immobilized mouse anti-HRV antibody at the test line (T) captures the complex, producing a visible red band (positive result).
