Dynamiker Gram Negative Bacterial Endotoxin Assay

It is used for quantitatively detecting Gram-negative bacterial Endotoxin (LPS) in dialysate, reverse osmosis water, replacement fluid, and other samples.

Endotoxin is a complex of lipopolysaccharide (LPS) and protein on the cell wall of Gram-negative bacteria. When the bacteria die or autolyze, endotoxin will be released. When a large amount of endotoxin enters the blood, it will cause a fever reaction - a "pyrogenic reaction".

Principle

Principle
  • The pre-treated samples are added into the Main Reagent which contains Factor C and Factor B. 

  • Factor C is activated by LPS and activated C activate Factor B, activated Factor B converts proclotting enzyme to clotting enzyme. 

  • The clotting enzyme hydrolyzes the substrate (Boc-leu-Gly-Arg-PNA) to release PNA. 

  • The absorbance is measured at 405nm kinetically. The concentration of LPS is interpreted according to a standard curve.

Procedure

Adaptation Platform

The ET Assay is working on a microplate reader with kinetic reading at 405nm and temperature control at 37℃.

Recommended microplate readers are Tecan Sunrise or BioTek ELX808iu, with incubator and kinetic reading at 405nm.
