The pre-treated sera are added to the main reagent containing Factor G. Factor G is activated by (1-3)-β-D-glucan and the activated Factor G converts the proclotting enzyme to the clotting enzyme.
The clotting enzyme hydrolyses the substrate(Boc-leu-Gly-Arg-PNA) to release PNA.
The absorbance is measured kinetically at 405 nm.
The concentration of (1-3)-B-D-glucan is interpreted from a standard curve.
The BDG Assay is working on a microplate reader with kinetic reading at 405nm and temperature control at 37C.
Recommended microplate readers are Tecan Sunrise or BioTek ELX808iu, with incubator and kinetic reading at 405nm.
